Jonathan is the son of wealthy businessman George Joestar I and Mary Joestar. He is an honest, kind and positive man whose life turns into a tragedy after meeting his adopted older brother - and the antagonist of the series - Dio Brando. In the fight against Dio, Jonathan becomes a user of Hamon under the guidance of his teacher Will Antonio Zeppeli. His legacy extends to almost all of the first six installments of the series, and his descendants include characters such as Joseph Joestar and Jotaro Cujo.

Jonathan Joestar's grandson, Joseph, is a natural user of Jamon and, eventually, a Booth user who owns a psychic photography booth, Hermit Purple. A violent delinquent with an exceptional talent for cunning, Joseph, with initiative and impressive ingenuity, confronts the fantastic threats that beset him throughout his life, fighting vampires, Pillar People, and enemy stand users. Joseph is the second most frequently appearing Jojo in the series after his grandson Jotaro Kujo. He is also the father of Josuke Higashikata, the main protagonist of the Indestructible Diamond.

Jotaro is a delinquent with Japanese roots on his father's side and semi-British, semi-Italian roots on his mother's side. Jotaro lives a normal life until the old sworn enemy of the Joestar family, DIO, returns. After seeing that his mother, Holy, has fallen ill (caused by DIO's awakening), Jotaro embarks on a journey around the world with his grandfather, Joseph Joestar, and a group of friends to save his mother and defeat the vampire once and for all. Possessing the all-powerful Star Platinum, Jotaro is the first JOJO presented with the stand and one of the most famous characters in the series. He is the ex-husband of an American woman and the father of Jolene Cujo, the main character of the Stone Ocean.

Josuke is a biracial first—year high school student living in the city of Morio. In 1999, he meets Jotaro Kujo, who informs him that he is Joseph's illegitimate son, making him a member of the Joestar family. After the death of his grandfather, Joske takes it upon himself to protect his beloved city from the evil owners of the stands. Josuke is a stand user with a Crazy Diamond stand that can restore almost anything.

As the illegitimate son of Dio Brando, conceived by the stolen body of Jonathan Joestar, Giorno Giovanna, first introduced as Haruno Shiobana, is a teenager living in Italy. He talks about his intention to join the powerful Passione gang and his dream of becoming a "Gangstar" in order to restore the virtue of the corrupt mafia. Giorno is a stand user with a natural background who possesses the powerful life source Gold Experience in battle.

Jolene Cujo is the main protagonist of the sixth installment of the Incredible Adventures of Jojo series, Stone Ocean and, accordingly, the sixth Jojo in the series. She is the daughter of an American woman and Jotaro Kujo, the protagonist of the Stardust Crusaders. Framed for a drunken murder, Jolene is sent to Green Dolphin Street Prison, where she investigates the case and battles Dio's most trusted student, Father Enrico Pucci. During her time in prison, Jolene awakens her stand, Stone Free, powered by strings.

Johnny Joestar is the main protagonist of the seventh installment of the Incredible Adventures of Jojo series, the Steel Ball Race, as well as the seventh JOJO in the series. He also appears as a posthumous minor character in Jojoleon. A former jockey turned paraplegic, Johnny participates in the Steel Ball race to find out the secret of Gyro Zeppeli's steel balls, because only they can heal his legs. During the race, Johnny becomes both a Spin user and a stand user when he acquires Tusk.

Josuke is a young man suffering from retrograde amnesia and devoid of any memories before he was discovered by Yasuho Hirose near the Seer Walls in the city of Morio. He dedicates himself to revealing his former identity and those who were originally associated with him. Josuke's original identity was Josefumi Kujo, but he became his current identity after merging with the body parts of Yoshikage Kira. Josuke is a user of the Soft & Wet stand and keeps it in its original form.
